Descripció del contingut i pla d'activitats:
Els canvis ambientals van lligats a canvis en la expressió dels
gens que permeten a les plantes aclimatar-se a la nova situació.
Aquests canvis transcripcionals poden ser regulats a nivell
epigenètic mitjançant el control de l'estructura de la cromatina.
Entre els diferents mecanismes epigenètics que controlen aquest
procés hi ha les modificacions post-traduccionals de les
histones. L'objectiu d'aquest projecte és relacionar determinades
marques d'histones amb els canvis d'expressió de certs gens quan
les plantes es troben en condicions d'estrès per altes
temperatures. Dades preliminars relacionen la trimetilació de la
lisina 27 de la histona H3 (H3K27me3) amb la resposta a estrès
tèrmic, doncs mutants del grup de les proteïnes Polycomb
(encarregades de depositar i estabilitzar la H3K27me3) tenen in
fenotip deficient en la resposta a altes temperatures (com per
exemple el mutant lhp1).
Per poder aprofundir en la possible relació entre l'activitat
Polycomb i l'estrès per altes temperatures, el projecte contempla
analitzar el fenotip a 28ºC (estrès) de diversos mutants
d'Arabidopsis deficients en membres del grup de les proteïnes
Polycomb. A nivell molecular, s'analitzarà mitjançant RT-qPCR
l'expressió de gens (entre ells HFR1) en condicions d'estrès i es
correlacionarà amb els nivells de H3K27me3 analitzats per
immunoprecipitació de cromatina (ChIP). A més a més, a partir de
dades de transcriptòmica i epigenòmica publicades, es
profunditzarà en la relació que pot haver entre l'activitat
Polycomb i l'estrès per altes temperatures.

Overview (resum en anglès): The chromatin remodeling protein LIKE HETEROCHROMATIN PROTEIN1 (LHP1) participates in the transcriptional regulation of a wide variety of genes. It is known that the mechanism for doing this is through Polycomb activity and is therefore related to epigenetic marks, specifically in the trimethylation of lysine 27 in histone H3 and the ubiquitination of histone 2A, H3K27me3 and H2Aub respectively. The role of LHP1 in the regulation of plant response genes to environmental changes is uncertain. The aim of this work is to study it, specifically in the response to high temperatures.
Through phenotype, gene expression and chromatin immunoprecipitation analyses, the response to high temperatures of Arabidopsis thaliana seedlings and the role of LHP1 have been analyzed using the lhp1 loss-of-function mutant. It has been observed that LHP1 plays an important role in the response of Arabidopsis seedlings to high temperature, as evidenced by the lack of hypocotyl elongation in its mutant. This lack of elongation is not due to a problem with auxin synthesis, as the phenotype of lhp1 in response to high temperatures could not be complemented by the addition of exogenous auxin. Therefore, the lhp1 mutant is insensitive to auxins, indicating that this phenotype is not due to a lack but to a deficiency in auxin signaling. In accordance with this, the mutant has a low expression of the IAA29 gene, a member of the Aux/IAA family of proteins, which are essential for auxin signaling. Interestingly, the low expression of IAA29 is not related to the levels of H3K27me3 and H2Aub, as these marks are neither present in this gene nor changed in the lhp1 mutant. Not finding a direct relationship between transcriptional activity and Polycomb activity, we used an lhp1 mutant whose LHP1 protein has lost its ability to bind to chromatin, we observe that the function of LHP1 in the regulation of response to high temperatures does depend on its binding to chromatin. This study opens the door to future work to find out how LHP1 regulates at the chromatin level the expression of the IAA29 gene and subsequently participates in the regulation of the plants' response to high temperatures.
