Descripció del contingut i pla d'activitats:
En els darrers anys s'ha detectat un augment preocupant de la
incidència de l'antracnosi en fulla i fruit de pomeres de
Girona. Fins ara, la incidència en fulla ja era coneguda, però
l'afectació del fruit és recent i representa un greu problema
per a la comercialització de la fruita. L'objectiu del
projecte actual és identificar i caracteritzar les espècies de
Colletotrichum, agent causal de l'antracnosi, associades a
aquesta malaltia de les pomeres de Girona. La identificació i
caracterització dels fongs seguirà una doble vessant: 1)
l'estudi de caràcters morfològics dels fongs, usats en la
identificació taxonòmica tradicional, i 2) la seqüenciació de
l'ADN dels fongs i posteriors anàlisis filogenètiques per a la
determinació dels tàxons.

Overview (resum en anglès): Anthracnose, caused by species of the genus Colletotrichum, is one of the most economically important fungal diseases of apple worldwide. The aim of this study was to identify and characterise the Colletotrichum species associated with anthracnose outbreaks detected in commercial apple orchards in the Baix Empordà region (Girona, Spain). Twenty-one Colletotrichum isolates were obtained from fruit (n = 20) and leaf (n = 1) samples of the cultivar 'Pink Lady' collected from two commercial orchards in Gualta (Girona). Morphological characterisation included the study of colony growth and the morphology of conidia and appressoria. For molecular characterisation, six genetic regions (ITS, GAPDH, ACT, CAL, TUB2 and ApMAT) were sequenced and a multilocus phylogenetic analysis was performed using Bayesian inference. Morphological characterisation revealed homogeneous morphology among all isolates, consistent with the C. gloeosporioides species complex, but insufficient for species-level identification. Molecular characterisation confirmed that all isolates belong to Colletotrichum chrysophilum, showing 99-100 % similarity to reference sequences in GenBank and maximum posterior probability (PP = 1.00) in the Bayesian phylogenetic tree. This is the first report of C. chrysophilum as the causal agent of apple anthracnose in Spain, confirming the spread of this emerging pathogen in Europe. The results highlight the need for multilocus molecular identification strategies for the accurate diagnosis of Colletotrichum species.
