Descripció del contingut i pla d'activitats:
A Catalunya, el nematode fitoparàsit que causa les majors pèrdues de producció en cultius hortícoles és el nematode formador d'agalles, Meloidogyne spp. Les espècies més abundants a casa nostre són M. incognita, M. javanica i M. arenaria. A causa de l'augment de temperatures es preveu un desplaçament d'aquestes cap a zones hortícoles del nord d'Europa. No obstant això, aquestes per poder desenvolupar-se hauran de pasar periodes freds a l'hivern.
Per tal de veure si aquestes espècies desplaçaran les actuals al nord d'Europa, es vol amb aquest projecte, determinar la supervivència a baixes temperatures del Meloidogyne spp.
Per assolir aquest objectiu, es realitzaran dos experiments, un amb ous i un altre amb formes juvenils. Es col·locaran pots amb sòl esterilitzat mesclat amb ous o juveniles del nematode a 3 temperatures (-3ºC, 0ºC i 3ºC). Un cop passat trenta dies. El sòl es col·locarà en testos i es col·locarà una tomaquera. Al cap de 40 dies es determinarà la possible reproducció.

Overview (resum en anglès): Meloidogyne incognita, M. javanica, and M. arenaria are three of the most widespread root-knot nematode species worldwide, with an optimal temperature range of 20-25°C. Climate change could favor their establishment in colder climate zones where they have not currently become established. The aim of this study was to evaluate the survival of these three species at three low temperatures (3°C, 0°C, and ¿3°C), under the hypothesis that their survival would decrease drastically as temperature decreased. To do so, a combined inoculum of the three species was subjected to one month of continuous exposure to each temperature in closed pots with sterile sand, following a gradual acclimation pretreatment. Tomato seedlings were subsequently transplanted into the same pots, and after eight weeks, infection and nematode reproduction were assessed through the number of egg masses, the number of eggs, and fertility (eggs/mass), as well as the survival of the three species through juvenile identification by PCR. Prior to this, a viability trial was conducted comparing one day of inoculum storage at 4°C versus room temperature (RT), with species identification by PCR. The results confirmed the hypothesis: at 0°C and ¿3°C, total juvenile mortality occurred, with no evidence of infection, whereas at 3°C the three species maintained their infective and reproductive capacity. In the viability trial, the 4°C treatment produced more egg masses and more total eggs than the RT treatment, although with lower fertility per mass, and PCR showed clear differences in tolerance between species. Overall, this study confirms that Meloidogyne spp. juveniles do not survive at temperatures at or below 0°C maintained for one month, placing their real survival threshold between 0°C and 3°C.
