Descripció del contingut i pla d'activitats:
LHP1 és una proteïna clau en la regulació de la cromatina i contribueix a determinar quins gens s'activen i quins romanen silenciats. Mitjançant estudis previs s'ha identificat un conjunt de proteïnes que podrien interactuar físicament amb LHP1 i participar en els seus mecanismes de regulació. El repte ara és comprovar si aquestes interaccions són reals i directes dins la cèl·lula.
En aquest projecte, l'objectiu és determinar si LHP1 interacciona amb les proteïnes candidates mitjançant assajos d'expressió en Nicotiana benthamiana, una planta molt utilitzada per estudiar interaccions proteiques de manera ràpida i eficient. Per fer-ho, s'introduiran a les fulles per agroinfiltració construccions d'ADN que permeten expressar LHP1 i les proteïnes candidates amb diferents etiquetes que faciliten la seva detecció.
L'estudiant aprendrà el procés del clonatge i dues aproximacions complementàries per a l'estudi d'interacció proteïna-proteïna: la primera és la co-immunoprecipitació, una tècnica que permet "pescar" LHP1 de l'extracte proteic i comprovar si les proteïnes candidates s'hi troben unides; i la segona és la complementarietat de fluorescència (BiFC), en què LHP1 i la proteïna candidata s'uneixen a dues meitats d'una proteïna fluorescent: si interaccionen, la fluorescència es recupera i es pot observar al microscopi confocal.
El treball de laboratori es realitzarà mitjançant l'ús de llibreta electrònica connectada al núvol, i seguint les normes d'anotació establertes al grup. A part del treball experimental, l'estudiant participarà en les reunions setmanals de grup. També hi haurà la possibilitat de participar en els seminaris científics organitzats setmanalment per diverses institucions al campus de la UAB sobre bioquímica, biologia molecular i/o biologia vegetal.

Overview (resum en anglès): The 26S proteasome is the major proteolytic complex responsible for maintaining protein homeostasis in eukaryotic cells and plays a key role in plant responses to environmental stress. The genes PBE1 and PBE2 genes play an essential role in the proteasome, as they encode catalytic subunits responsible for the proteolytic activity of the proteasome. However, both genes are highly similar and present functional redundancy. The objective of this work was to investigate the role of PBE1 and PBE2 during high-temperature stress in Arabidopsis, through phenotypic and gene expression analyses, while generating molecular tools for their constitutive overexpression.
Phenotypic characterization revealed that, under control conditions (22 °C), the seedlings of pbe1 and pbe2 mutants were indistinguishable from wild-type plants, supporting the functional redundancy of these genes during normal growth. In contrast, prolonged exposure to 37 °C resulted in a significant increase in hypocotyl elongation in all genotypes, with both mutants exhibiting a stronger response than the wild type, indicating that PBE1 and PBE2 contribute to plant adaptation to prolonged heat stress. Expression analysis further revealed differential transcriptional regulation of the two genes: PBE1 maintained constitutive expression throughout the treatment, whereas PBE2 expression decreased significantly after 24 h of heat stress. These results suggest that, despite their structural similarity, PBE1 and PBE2 have undergone partial functional divergence and may differentially regulate proteasome activity during heat stress.
In parallel, Gateway-compatible overexpression constructs carrying PBE1 or PBE2 merged to the gene that codifies for the reporter protein FLAG were successfully generated, validated by Sanger sequencing and restriction analysis, introduced into Agrobacterium tumefaciens, and used for the stable transformation of Arabidopsis thaliana. Although the characterization of the transgenic lines was beyond the scope of this study, these molecular tools provide a valuable resource for future functional analyses of PBE1 and PBE2 in plant stress responses.
