El laboratori Protein Design and Modeling del Institut de Biologica Molecular de Barcelona que treballa en el disseny computacional de proteïnes. Un cop s¿identifiquen seqüències d'interès, aqueste son expressades de manera recombinant per caracteritzar-ne les propietats bioquímiquesi estructurals.
La idea al TFG és provar mètodes recents basats en intel·ligència artificial per identificar imperfeccions de dissenys que després deriven en problemes d'expressió, formació d'oligòmers no desitjats o errors importants en la seva estructura quan s'ha resolt posteriorment.
Provarem aquests mètodes per optimitzar proteïnes dissenyades al nostre laboratori, i si s'obtenen resultats prometedors llavors comprovar-les experimentalment.

The computational de novo protein design has taken a drastic upswing due to the accessibility of new procedures such as the ones given by Artificial Intelligence (AI). This method not only helps with the prediction and comprehension of the geometry of the proteins but opens the door to new personalised biomedical and biological engineered-drugs. In this thesis I aim to adapt a script to test the possibility for optimising a design of a protein previously checked experimentally. Specifically, dIG14, a design of an immunoglobulin-like (Ig) ß-sandwich scaffold without compromising its conformation and investigating the broad range response given by this AI method. This approach tries to enhance the solubility of the Ig surface and the reconstruction of its structure analysed with the prediction-by-sequence techniques. The outcome is restricted and validated into certain values of protein computational analysis (RMSD or pLDDT) and is further pursued by structure prediction techniques leading to a few possible models remarking the reduction of hydrophobicity and dimerization. I finally obtained 14 different sequences capable of providing better stability fulfilling all the previous requirements. The results show a clear representation of the versatility of these new methods and the possibility of utilising this programme for the optimization of determined regions of proteins.
